Background The USDA Agricultural Research Service (ARS) National Cold Water Aquaculture Research Facility (NCCCWA) is issuing this solicitation for the purpose of conducting protein analysis on rainbow trout. The goal of this contract is to obtain unbiased quantitative proteomics analysis to support research efforts in aquaculture. Work Details The contractor will perform 4D-DIA (data independent acquisition) on a LC-MS/MS platform for 36 lysosomal protein preparations isolated from rainbow trout liver. The work includes: 1. Sample preparation involving protein extraction, denaturation, reduction, tryptic digestion, and peptide cleanup. 2. Establishment of a spectral library using a pooled sample. 3. DIA data collection must be conducted using a timsTOF mass spectrometer (LC-MS/MS). Deliverables include a technical report detailing the experimental procedure, LC-MS/MS instrument parameters, and all identified and quantified data. The analysis should allow for pairwise comparisons between treatment groups and provide data in an Excel spreadsheet format within 60 days of sample receipt.
